A model/hero with Holy can reroll an atk die against an undead model for instance the paladin. This is done before the undead model makes it's defense roll correct? not after.

A hero with Holy is not the same as a hero that attacks with an Holy attack correct? For instance the say the nun gives the barb her potion to allow him to do cross 1, holy, and fire. The Barb now attacks an undead model. He does not apply the reroll correct? as it is not an his ability. It's only a type of damage inflicted.

You're correct, the re roll would be taken before the defense roll is made.

The sister of light's potion (elixir of light) would grant any hero who drinks it the "holy" effect so they would get the re roll against undead and demons for the rest of their activation.

It's also worth noting that elixir of light gives cross 1 to the hero who drinks it. If the cross effect targets multiple monsters one of whom is undead/ demon type you would get the re roll on the offensive roll even if it is only targeting one undead monster and say 3 regular monsters
As per the FAQ thread in the rules forum,
Q - How do type bonuses (Holy vs Undead) affect an AOE roll when there are also mobs of a different type? Do you get to reroll for everyone, just that type, or no roll at all ?
A - If a model within the AoE is of the affected type (e.g. Undead) the special ability is triggered, and applies to all models within the AoE regardless of their type.
